For travel from Prague Airport, we recommend using Gepard Express (link), which offers direct buses from the airport to Brno. The journey takes about 2.5 hours.
Alternatively, you can use RegioJet (link), which also provides buses, but you need to transfer once at Prague’s main bus station, Florenc. This makes the trip longer—about 4 hours. However, RegioJet has more frequent departures than Gepard Express.
